چکیده

The purpose of this experimental work was to determine whether the precancerous loss of rat liver 5-hydroxytryptophan (5-HT) decarboxylase was associated with sequential atrophic alterations preceding hepatocarcinogenesis. To test whether such an association existed, three experimental conditions were employed in which hepato carcinogenesis was either markedly delayed or prevented. Under all three experimental conditions the losses of 5-HT decarboxylase activity approximated those observed in precancerous livers of rats fed 3'-methyl-4-dimethylaminoazobenzene. It was also shown that the losses were not associated with proliferation stimulated by partial hepatectomy. It was concluded that the precancerous loss of 5-HT decarboxylase activity either (a) was associated with factors other than those involved with sequen tial atrophic alterations preceding hepatocarcinogenesis or (b) if associated with the atrophic alterations, was a necessary but insufficient reaction leading to malignant transformation. The possible correlation between the activity losses and protein-bound azo dyes was discussed.

The effect of hepatocarcinogenesis upon 5-hydroxytryptophan decarboxylase and serotonin deaminase.

Enzymes associated with the catabolism of 5-hydroxytryptophan (5-HT) were assayed in transplanted hepatomas, in primary lesions induced with 8'-methyl-4dimethylaminoazobenzene (St-Me-DAB), and in 8'-Me-DAB precancerous rat liver. Among six transplanted rat hepatomas, 5-HT decarboxylase activity was essentially absent in five, whereas the Morris hepatoma 51~8 manifested activity equivalent to th...

Effect of pyridoxal phosphate deficiency on aromatic L-amino acid decarboxylase activity with L-DOPA and L-5-hydroxytryptophan as substrates in rats.

This paper describes the distribution of aromatic L-amino acid decarboxylase (AADC) activities in fourteen tissues (eight peripheral tissues and six brain regions) of semicarbazide (SC)-treated rats, using both L-DOPA and L-5-hydroxytryptophan (L-5-HTP) as substrates. The distribution of pyridoxal phosphate (PLP) was also measured in control and SC-treated rats. SC-treatment decreased the PLP c...

On the identity of DOPA decarboxylase and 5-hydroxytryptophan decarboxylase (immunological titration-aromatic L-amino acid decarboxylase-serotonin-dopamine-norepinephrine).

Simultaneous immunological titration of DOPA and 5-hydroxytryptophan decarboxylase activities, from a number of tissues of various species, showed that the two activities were not distinguishable with a monospecific antiserum to hog kidney decarboxylase. Together with previous findings, these data firmly establish the concept that in mammalian tissues the two enzyme activities are associated wi...

DOPA Decarboxylase Modulates Tau Toxicity

BACKGROUND The microtubule-associated protein tau accumulates into toxic aggregates in multiple neurodegenerative diseases. We found previously that loss of D2-family dopamine receptors ameliorated tauopathy in multiple models including a Caenorhabditis elegans model of tauopathy. METHODS To better understand how loss of D2-family dopamine receptors can ameliorate tau toxicity, we screened a ...

A simple determination of serotonin, 5-hydroxyindoleacetic acid and 5-hydroxytryptophan decarboxylase activity in rat brain areas and parallel correlation among the levels.

We devised a simple method for the separation of serotonin (5HT) and 5-hydroxyindoleacetic acid (5HIAA) using small P-cellulose and DEAE-Sephadex columns, respectively. Both substances were estimated fluorometrically by the reaction with o-phthalaldehyde (OPA). In this method, some known interfering substances in addition to many primary amines and amino acids were separated from 5HT and 5HIAA....
